Soon i will buy the Zwift Kickr core one coq. I know that the coq is pre installed.
My bike is mechanical shifting 10 speed. For the the smoothest sound i must shifting front on the lowest en behind on the 5 or 6.
But when must i shifting ? when my weel is out and my chain is on the one coq ? or before?
Either is fine. You can shift it with wheel on or off.
Having it in the middle should make it easier to place onto the cog but it will soon become obvious if it needs shifting you can just tap the gears as you fit to the core.

Sorry for the delay, but I couldn’t miss the chance to help you in this matter. I want you to know that we recommend that you shift your bike’s gears so the chain is on the smallest chain ring on the front and one of the middle cogs at the back before you get your bike on the Zwift Cog and after your wheel is out. For instance, if you have 10 gears, shift into gear five or six.
Once you do this you can move forward mounting your bike on the Zwift Cog, and sort out your chain line by shifting up or down to align the derailleur! You can get more visual guidance from this video.
